Studies on Embryo Culture of Peaches (Prunus persica)



Abstract
Factors affecting in vitro organ differentiation of early-ripening Prunus persica (cv. Kuraliatawase) embryos are described.
Brooks & Hough medium with 20g/§¤ sucrose was the best medium, and addition of sucrose was essential for the normal shoot growth. Harvesting the fruits 5 to 10 days before optimal harvest date (OHD) was better than harvesting on OHD for the organ differentiation of embryo and normal shoot growth. The percentage of embryo developing normal shoot was increased with the low temperature storage period passing. Percentage of germination was decreased, and contamination was increased significantly when the embryos were incubated with testa. Cotyledon was essential for the normal shoot growth, but partial removal of the cotyledon improved shoot and radicle length and shoot fresh weight.
